I have never ever switched off the ignition (or power, in the case of
machine tool controls or drive amplifiers, or anything else for that
matter) doing any kind of voltage checks.  And I know that the team of
CNC machine tool service techs that worked for me didn't either. 
Current checks maybe, depending.  But voltage checks, no.
